Insulin acts as a glucagon release-inhibiting paracrine hormone, especially at high concentrations. So then the body releases cortisol. Cortisol comes to the brain’s rescue in 2 ways: 1- Increases gluconeogenesis, the making of glucose by breaking down proteins from skin, muscle and organ tissue and converting them to glucose in the liver. 2- Causes a moderate decrease in the rate of glucose utilisation by cells everywhere in the body (cortisol causes insulin resistance [IR]). So, the first 3 pillars of ageing excess insulin, cortisol and blood glucose are all interlocking and mutually enhancing.

Endogenous levo-carnitine (β-hydroxy-γ-trimethylammonium butyrate), found in many human tissues, is an amino acid derivative. It is synthesised in the liver and kidney from proteinbound lysine (oral lysine cannot improve carnitine synthesis) and methionine. It is a watersoluble compound that exhibits biologic activity only when in the levo isoform. Several enzymes and cofactors (iron, ascorbic acid, niacin, and pyridoxine) are involved in its biosynthesis, and only one matrix mitochondrial enzyme is involved in the pathway.

Insulin lowers the concentration of glucose in blood by inhibiting hepatic glucose production and by stimulating the uptake of glucose by muscle and adipose tissue. Insulin inhibits lipolysis, stimulates fatty acid synthesis and decreases the hepatic concentration of carnitine FAT AND MUSCLE DYSFUNCTION 47 (carnitine shuttles fatty acids into mitochondria in most cells for use as ATP energy fuel). Insulin stimulates the fat cells to take up fat and sugar from the blood and store it away as body fat, especially in the middle of the body, within the abdomen and around the vital organs.
